A Substrate for Uniform Production of Perithecia In Gibberella Zeae

Abstract
SUMMARY Fresh carnation leaf tissue treated with ethanol offers a satisfactory substrate for the study of sexual reproduction in Gibberella zeae. Carnation leaf tissue disks were soaked in 70% aqueous ethanol for 5 min, thoroughly rinsed in distilled water, and placed on water agar. Following inoculation of leaf disks with macroconidia and incubation for 10 days, uniform, reproducible numbers of perithecia were formed. Perithecia produced on this substrate discharged ascospores. The method is suitable for assaying the effect of substrate and environmental factors on sexual reproduction.
