The novel coronavirus SARS-CoV-2 is an acute respiratory tract infection that emerged in Wuhan city, China. The spike protein of coronaviruses is the main driving force for host cell recognition and is responsible for binding to the ACE2 receptor on the host cell and mediates the fusion of host and viral membranes. Recognizing compounds that could form a complex with the spike protein (S-protein) potently could inhibit SARS-CoV-2 infections. The software was used to survey 300 plant natural compounds or derivatives for their binding ability with the SARS-CoV-2 S-protein. The docking score for ligands towards each protein was calculated to estimate the binding free energy. Four compounds showed a strong ability to bind with the S-protein (neohesperidin, quercetin 3-O-rutinoside-7-O-glucoside, 14-ketostypodiol diacetate, and hydroxypropyl methylcellulose) and used to predict its docking model and binding regions. The highest predicted ligand/protein affinity was with quercetin 3-O-rutinoside-7-O-glucoside followed by neohesperidin. The four compounds were also tested against other related coronavirus and showed their binding ability to S-protein of the bat, SARS, and MERS coronavirus strains, indicating that they could bind and block the spike activities and subsequently prevent them infection of different coronaviruses. Molecular docking also showed the probability of the four ligands binding to the host cell receptor ACE2. The interaction residues and the binding energy for the complexes were identified. The strong binding ability of the four compounds to the S-protein and the ACE2 protein indicates that they might be used to develop therapeutics specific against SARS-CoV-2 and close related human coronaviruses.

Recently, the application of molecular docking is drastically increasing due to the rapid growth of resolved crystallographic receptors with co-crystallized ligands. However, the inability of docking softwares to correctly score the occurred interactions between ligands and receptors is still a relevant issue. This study examined the Pearson’s correlation coefficient between the experimental monoamine oxidase-B (MAO-B) inhibitory activity of 44 novel coumarins and the obtained GOLD 5.3 docking scores. Subsequently, optimization of the docking protocol was carried out to achieve the best possible pairwise correlation. Numerous modifications in the docking settings such as alteration in the scoring functions, size of the grid space, presence of active waters, and side-chain flexibility were conducted. Furthermore, ensemble docking simulations into two superimposed complexes were performed. The model was validated with a test set. A significant Pearson’s correlation coefficient of 0.8217 was obtained for the latter. In the final stage of our work, we observed the major interactions between the top-scored ligands and the active site of 1S3B.

SARS-CoV-2, a new type of Coronavirus, has affected more millions of people worldwide. From the spread of this infection, many studies related to this virus and drug designing for the treatment have been started. Most of the studies target the SARS-CoV-2 main protease, spike protein of SASR-CoV-2, and some are targeting the human furin protease. In the current work, we chose the clinically used drug molecules remdesivir, favipiravir, lopinavir, hydroxychloroquine, and chloroquine onto the target protein SARS-CoV-2 main protease. Docking studies were performed using Arguslab, while Discovery Studio collected 2D and 3D pose views with the crystal structure of COVID-19 main protease in complex with an inhibitor N3 with PDB ID 6LU7. Computational studies reveal that all ligands provided good binding affinities towards the target protein. Among all the chosen drugs, lopinavir showed the highest docking score of -11.75 kcal/mol. The results from this molecular docking study encourage the use of lopinavir as the first-line treatment drug due to its highest binding affinity.

Corona Virus Disease 2019 (COVID-19) is a new strain of coronavirus called SARS-CoV-2, which was identified in Wuhan, China, in December 2019. The rapid transmission of COVID-19 from human to human forced researchers to find a potent drug by setting aside the time-consuming traditional method in drug development. The molecular docking approach is one a reliable method to screening compound from chemical drug or by finding a compound from Indonesian herbal plants. The present study aimed to assess the potency of compounds from five medicinal plants as potential inhibitors of PLpro and 3CLpro from SARS-CoV-2 using molecular study. The molecular docking was performed using Protein-Ligand Ant System (PLANTS) to analyze the potential compounds by the docking score. Remdesivir triphosphate was used as a standard for the comparison of the test compounds. The docking score obtained from the docking of PLpro with native ligand, remdesivir triphosphate, curcumin, demethoxycurcumin, bisdemethoxycurcumin, luteolin, apigenin, quercetin, kaempferol, formononetin-7-O-glucuronide, andrographolide, and neoandrographolide were -111.441, -103.827, -103.609, -102.363, -100.27, -79.6655, -78.6901, -80.9337, -79.4686, -82.1124, -79.1789, and -97.2452, respectively. Meanwhile, docking score with 3CLpro for the same ligand were -64.0074, -86.1811, -81.428, -87.1625, -78.2899, -73.4345, -70.3368, -71.5539, -68.4321, -72.0154, -75.9777, and -93.7746. The docking score data suggest that curcumin was the most potential as a PLpro inhibitor, while neoandrographolide was the best as a 3CLpro inhibitor.

The systemic lupus erythematosus (SLE), commonly known as Lupus, is a rare and complex multisystem autoimmune disease where one’s immune system is overactive, and the body attacks its organ systems. SLE is a historically old disease described already in antiquity; it is an example of a chronic disease with physical, psychological, financial, and social implications for individuals diagnosed. It has inspired medical and basic biological scientists that focus on molecular biology, basic immunology, immunopathology, clinical science, genetics, and epidemiology. The syndrome is real in its existence-although hidden behind obstacles, cumbersome for patients and clinicians, and rebellious for scientists. There is currently no cure for SLE. The goal of treatment is to ease symptoms. This article will review information on the general approach to SLE therapy, focusing on currently approved therapies and novel approaches that might be used in the future.

This study aims to develop a liposomal drug delivery system of isotretinoin, an acne drug-using spray drying, as a cost-effective and time-effective technique. The liposomal formulation was prepared by using spray drying; three different strategies were adopted: suspension spray drying (SSD), thin-film hydration and spray drying (TFHSD), and emulsion spray drying (ESD). Isotretinoin was 99% bound with lipid, so lipids hydrogenated soy phosphatidylcholine (HSPC), distearoyl phosphatidylglycerol (DSPG), and cholesterol were selected for the formulation development. The HSPC, DSPG, cholesterol, and isotretinoin were taken in the ratio 4 : 1 : 0.16 : 3.1 mmol. In vitro drug release studies, microscopy, drug content, and related substance characterizations were done to formulate each strategy of spray drying prepared dry liposomes of isotretinoin. Results were compared with the USP monograph of isotretinoin. It was revealed that isotretinoin's liposomal formulation using ESD was having drug release according to the USP limits. Drug content was also according to the USP requirement; no free drug crystals were found in microscopy, multivesicular vesicles were found in shape, a particle size of up 60 µ was found. The ESD technique was a successful, time-effective, and cost-effective technique for preparing a liposomal drug delivery system for isotretinoin.

Glimepiride is often combined with metformin HCl as an oral antidiabetic in type II diabetes mellitus, which provides a complementary and synergistic effect with multiple targets for insulin secretion. Glimepiride includes class II of BCS, which solubility practically insoluble in water but high permeability, which will impact the drug's small bioavailability. In contrast, metformin HCl includes class III of BCS, which has a high solubility in water, but low permeability is absorbed approximately 50-60% in the digestive tract given orally. The co-crystallization method can be used to improve the glimepiride solubility properties and the permeability properties of metformin HCl by interrupting glimepiride with metformin HCl physically. This study aims to identify the physical interactions between glimepiride and metformin HCL using a thermal analysis of Differential Scanning Calorimetry (DSC) and then confirmed by a computational approach. Identifying the physical interactions between glimepiride and metformin HCL was carried out by plotting the melting points generated from the endothermic peaks of the DSC thermogram at various compositions versus the mole ratios of the two were further confirmed by the computational approach using PatchDock. The results of the phase diagram analysis of the binary system between glimepiride and metformin HCl show a congruent pattern, which indicates the formation of co-crystal or molecular compounds at a 1 : 1 mole ratio at 228°C. Computational approach results showed that the interaction between glimepiride and metformin HCl did not form new compounds but heterosinton formation that was stable in molecular dynamics simulations.

Surgical site infections (SSIs) are challenging to treat and often associated with much higher extended stays, morbidity, and mortality, higher treatment costs, especially when the causative agent is multidrug resistance (MDR). This study was designed to determine the prevalence of nosocomial infections and susceptibility profiles of bacteria isolated from Cesarean section (C-section) and surgical patients from Kasese District Hospitals in Western Uganda. A descriptive cross-sectional study was conducted from January to September 2016 involving 303 patients with SSIs in obstetrics & gynecology; and general surgery wards in three health facilities. Clinical-demographic characteristics of patients were obtained using structured questionnaires before surgery. Bacterial analysis of the air and floor of the theatre room was done using the standard culture method. Of the 303 patients enrolled with SSIs (median age 34 years), 71.6% were female, and 28.4% were males. Only 14.5% developed SSIs, with predominant isolates being Staphylococcus aureus 33.33% and Escherichia coli 24%. The majority of recruited participants underwent a C-section of 58% and the least amputations of 0.3%. Duration of operation or surgery, p-value 0.002 (95% CI 1.599-7.667) was significantly associated with SSIs. Gram-negative bacteria were found resistant (50-100%) to ampicillin, gentamycin, and ciprofloxacin, the commonly used post-operative drugs of choice. Hospital-acquired infections were common with emerging antibiotic-resistant strains isolated in most SSIs at Kasese hospitals. The development of resistance to commonly used antibiotics such as ampicillin, gentamycin, and ciprofloxacin than previously reported calls for laboratory-guided SSIs therapy and strengthening infection control policies.

Drug-related problems (DRPs) had often been a concern in the system that needed to be detected, avoided, and addressed as soon as possible. The need for a clinical pharmacist becomes even more important. He is the one who can not only share the load but also be an important part of the system by providing required advice. They fill out the patient's pharmacotherapy reporting form and notify the medical team's head off any drug-related issues. General practitioners register severe adverse drug reactions (ADRs) yearly. As a result of all of this, a clinical pharmacist working in and around the healthcare system is expected to advance the pharmacy industry. Its therapy and drugs can improve one's health quality of life by curing, preventing, or diagnosing a disease, sign, or symptom. The sideshows, on the other hand, do much harm. Because of the services they offer, clinical pharmacy has grown in popularity. To determine the overall effect and benefits of the emergency department (ED) clinical pharmacist, a systematic review of clinical practice and patient outcomes will be needed. A clinical pharmacist's anatomy, toxicology, pharmacology, and medicinal chemistry expertise significantly improves a patient's therapy enforcement. It is now important to examine the failure points of healthcare systems as well as the individuals involved.

This study aimed to improve the dissolution rate of aceclofenac and release the drug in a controlled manner over a period of 24 hours. Matrix tablets were prepared by direct compression method, using hydrophilic polymers (HPMC/guar gum). Matrix tablets were prepared by wet granulation method using different hydrophilic polymers (HPMC/guar gum). Tablets were evaluated for in vitro drug release profile in phosphate buffer with pH 6.8 (without enzymes). The thickness and hardness of prepared tablets were 3.23 ± 0.035 to 3.28 ± 0.008 mm and 3.26 ± 0.115 to 3.60 ± 0.200 kg/cm2, respectively. The friability was within the acceptable limits of pharmacopoeial specifications (0.31 to 0.71%), which indicates the good mechanical strength of the tablets. Drug release was retarded with an increase in polymer concentration due to the gelling property of polymers. The in vitro drug release from the proposed system was best explained by Higuchi’s model, indicating that drug release from tablets displayed a diffusion-controlled mechanism. The results clearly indicate that guar gum could be a potential hydrophilic carrier in developing oral controlled drug delivery systems. Based on the study results, formulations F8 was selected as the best formulation.

Cod liver oil (CLO) has long been used as medicine or as a functional food. The CLO is a potential source of vitamin D, vitamin A, and omega fatty acids (eicosapentaenoic acid/EPA and docosahexaenoic acid/DHA). Self-nanoemulsifying drug delivery system (SNEDDS) can enhance dissolution, absorption, and bioavailability of hydrophilic and lipophilic substances for oral administration. The objective of this study was to develop a SNEDDS of CLO with good physical characteristics and stability. The optimization formula was carried out using various ratios of oil, surfactant, and cosurfactant. The physical properties of SNEDDS were determined by transmittance percentage, dispersibility, robustness, thermodynamics stability (heating-cooling cycle, centrifugation, and freeze-thaw cycle), and globule size distribution. The optimum formula of CLO-SNEDDS was obtained at a ratio of surfactant and cosurfactant 2 : 1 and a comparison of oil and surfactant mixtures 1 : 6. The CLO-SNEDDS meets the requirement of percent transmittance (97.90±0.85), dispersibility (grade A), and stability based on robustness and thermodynamic stability tests. Diluted SNEDDS has an average globule size of 125 nm with a polydispersity index (PDI) of 0.515. CLO-SNEDDS preparation has good physical characteristics and stability.

Food is a basic need for humans that must be fulfilled as a basic need; food has an essential meaning and role in a nation's life. The problems faced during the Covid 19 pandemic have forced everyone to keep quiet and work from home so that everyone tries to take advantage of this free time with productive activities. One of these activities is gardening at home using household waste. To make this happen, from April to September 2020, the community service team conducted counseling and training and practices for the Amuktiwo Ngubo farmer group, Kestalan sub-district, Banjasari District, Surakarta City, Central Java. The results of the program show that the Covid 19 pandemic forces every citizen, especially partner groups, to do activities at home, which results in a lot of free time available, so gardening at home by using household waste as a planting medium is the most appropriate solution because the environment is clean and healthy. The crop yields can meet the needs of the kitchen, which results in reduced expenses. Also, if this program is implemented nationally, the goal of community food security will be achieved.

Cawas Village has launched a Garbage Bank in every RW in its area, one of which is RW 09 Kuncen, which has succeeded in establishing the Manunggal Sejahtera Waste Bank. The focus of this program is the development of a waste bank to become the Parent Garbage Bank, both in management and in technical implementation. The objective of this activity is the compilation of the Cawas Village Kuncen Waste Bank Master Development Plan which is in line with the Cawas Village RPJMDes regarding waste management, both inorganic and organic. The Direct Action method is applied in community service programs by increasing management capabilities through socialization activities, training, practice, study visits, and Waste Bank Master planning documents preparation. The presence of the Covid-19 pandemic diverted some community empowerment activities to community efforts in overcoming Covid-19. Community empowerment is done online. The results achieved have not been optimal due to the Covid-19 pandemic, so that several strategic activities related to the initiation of the main waste bank development have been hampered. Some of the results were achieved, the agreement to build a waste bank was continued after the pandemic, a demonstration area for the reuse of waste as a planting place, preparation and implementation of an environmental competition for waste management, and activities to prevent the spread of Covid-19 (environmental sanitation and checking body temperature at the entrance to villages and mosques). The response of partner satisfaction to program implementation is above 80%, and 100% hope that it will continue

The problems faced by coconut shell charcoal craftsmen are production capacity, product quality, and low selling prices. Low product quality is due to the absence of quality standards in the production process. This activity aims to improve the management quality and add value to charcoal products by turning them into briquettes to increase the selling price. Implementing activities is divided into two different problem areas that one partner handles: production problems and management problems (raw material management, production process management, packing, and warehousing management, shipping management, product quality assurance management, and marketing management). The output of this community service activity is that coconut shell charcoal has been produced with an increased quality than before in terms of the low water content of less than 7%. Likewise, the briquettes to those produced are of high quality. There is an increase in added value through processing shell charcoal into briquettes which have significantly increased revenue and profit.

Dukuh Sikepan Ngagrong Village is one of the villages located in Gladagsari District, Boyolali Regency, Central Java Province. Plantations and animal husbandry are the sources of livelihood for the people of Dukuh Sikepan, most of whom work as farmers and breeders. Also, the limited knowledge of residents about animal feed fermentation has made residents only provide feed with ordinary grass, which is only found in the rainy season. So that during the dry season, residents have to buy feed elsewhere at a higher cost. The not yet optimal marketing of agricultural products and development is also an obstacle for Sikepan residents. Thus, it is necessary to have a community service program that is expected to help the people of Sikepan Hamlet reduce the problems they are facing. Through the method of extension, education, and assistance to develop public knowledge to recognize and utilize agricultural knowledge about animal feed fermentation and education about post-harvest online marketing in Sikepan Hamlet, it is necessary to do it. KKN students, assisted by Field Supervisory Lecturers, carry out community service programs that interact with the community. The KKN-PPM program is beneficial for residents in carrying out livestock feed fermentation, providing technical training to enable farmers to do what they need. Besides that, with online marketing training, mothers and adolescents can sell their products smartly.

Based on interviews conducted with the Head of the Vocational High School Management and Business Vocational High School Subject Teacher Conference in West Sumatra Province, it was found that teachers need to improve their competence about the use of Computer Accounting Applications. It is known that teachers still lack competence, and teachers are still unfamiliar with how to operate various menus and functions that can be used in a spreadsheet program to complete company financial reports. Based on this condition, the teaching and learning process for subjects has been done more with the lecture method and a little practice. So that this affects the competence of SMK graduates. In fact, the guidance for the world of work requires SMK graduates to operate this application program properly. The community's activity was carried out for two days on 18 and 19 September 2020 at SMKN 2 Kota Padang. The implementation is carried out in three stages, namely material presentation, integrated training, and evaluation. At the end of the activity, it was known that there was an increase in the understanding of the participants regarding the preparation of financial statements of trading companies using Microsoft Excel with a case settlement value that could be achieved was 85.71%. Furthermore, this increase in teacher competence is expected to be transferred to students and is reflected in the value of UKK and the acceptance of graduates in the world of work.

This paper has the aim of demonstrating the importance of environmentally friendly educational props by utilizing used goods. The implementation of teaching and learning activities at Pre-Kindergarten Kota Yogyakarta has problems with limited educational aids (APE). The existence of APE is one of the keys in carrying out the learning process. The low level of teacher creativity in making APE, in this case, becomes the anchor to provide APE for the learning process. This APE can be made by enhancing the creativity between teachers and parents of parents. During the Covid-19 pandemic, teaching and learning activities in Pre-Kindergarten were carried out in their respective homes. To support the use of APE, training on the use of used goods was conducted. The village of Mentaren II is productive as the center of the snack processing industry. The majority of the people use agricultural land as a livelihood to make various snacks. The management of small industries in this village has been going on for a long time; business actors have their own materials and equipment in their processing practices. Pukades is one of the leading village programs in the direction of development in the village. With this superior village product, it is hoped that it can boost job opportunities in the countryside. So that systematically, it can increase purchasing power and reduce the level of poverty in the village. The sales prospect is quite developed, as evidenced by the many results of their complaining that they are traded in suburban stalls along the Kalimantan road, even directly with buyers. However, this industry's development has faced obstacles since the Covid-19 epidemic, which requires the public not to gather together. This has an impact, especially on business actors who have been relying on conventional marketing systems. At this stage, the community service team visits the village office and meets the village head and village officials, discussing village institutions' activeness, both BUMDes, and Posyantek, in the village. The service team immediately went into the field by visiting several houses that had businesses and conducting brief interviews with the procession of managing activities and various obstacles.

Samboja district in Kutai Kartanegara Regency has a fairly good economic potential: dragon fruit cultivation. Dragon fruit farmers in Samboja District, especially members of the women's farmer group in Sei Merdeka Village, face several problems. When they enter the harvest period, dragon fruit is attacked by plant pests. Dragon fruit is only a few and is supplied by middlemen who are also buyers of the dragon fruit harvest. Middlemen buy the dragon fruit at a meager price compared to market prices. On the other hand, farmers cannot sell dragon fruit themselves directly in supermarkets or in traditional markets. This PKM activity will provide training on how to plant dragon fruit, so that plant pests do not attack it to partner members of the women farmer groups and PKK women's groups. This effort can break the dependence of women farmer group members on middlemen. The PKK group of women who became partners in this activity was given knowledge and skills for further dragon fruit cultivation; the results of this cultivation were processed into various commercial food products, such as noodles, jam brownies, jelly, and syrup. The results and outputs of this PKM activity increase the knowledge and skills of members of the women farmer groups and PKK women in processing dragon fruit into various processed food products. It is also hoped that new MSMEs based on processed dragon fruit products will grow

Suggestions from researchers provide regular outreach to the community, with the aim to change the community mindset that is the main problem of this program. Not only socialization, the government should continue to evaluate and remind that this function continues to work by the Working Group.
