The effect of addition glutathione antioxidant on vitrification medium to the viability of Garut sheep (Ovis aries) oocytes

Abstract
Research has been conducted to investigate the viability of Garut sheep oocyte to evaluate the effect of glutathione antioxidants addition with a concentration of 0.5 mM, 1 mM, and 1.5 mM in the vitrification medium to the viability of Garut sheep oocytes. The oocytes was freezed at -196 °C for 7 days. A total of 135 oocytes first were matured in vitro with the addition of 1 mM glutathione for 24 hours. The oocyte were vitrified using 15 % ethylene glycol, 15 % dimethyl sulfoxide, 0.5 M sucrose and divided into four treatments group with the addition of 0 mM (KK), 0.5 mM (KP 1), 1 mM (KP 2), and 1.5 mM (KP 3) glutathione. Viability was evaluated through the luminescence produced by Hoechst and Propidium Iodide dyes. The percentage of viable oocytes are 69.70 % (0 mM), 72.73 % (0.5 mM), 76.47 % (1 mM), and 77.14 % (1.5 mM). The results tends to increase along with the addition of glutathione concentration. However, statistically, ANOVA were not significantly different among treatment groups (P > 0.05). The conclusion of the study is that the addition of glutathione antioxidants in the vitrification medium are able to maintain the viability of Garut sheep oocytes during vitrification, although the ANOVA test were not significant.This publication has 8 references indexed in Scilit:
