Abstract:
A study based on completely randomized design with three replications was carried out at Tuber Crops Research Centre (TCRC), Bangladesh Agricultural Research Institute (BARI), Gazipur with a view to evaluating and classifying agro-morphological traits in 12 potato varieties to enumerating the genetic diversity. Discriminant function analysis (DFA) was done to determine the set of discriminatory functions contributed to separating 12 potato varieties into four distinct clusters. Discriminant function analysis showed that, function 1 explained 78.21% and function 2 explained 21.79% of total variance. The first function was more related to leaf area (having coefficient value of 0.661) and second function was more related to minituber per plant and plant height (having coefficient value of 0.644 and 0.523, respectively). Among 4 clusters, cluster II showed the highest distance (29.79) with cluster III followed by the distance between clusters III and IV (24.70) so the varieties in these clusters were genetically diverse. Based on mean value of the six quantitative characters for each of the four clusters and the inter cluster distance the cluster III and IV might be selected for future breeding program as a better parent to get positive heterosis. Cluster III contained six varieties BARI Alu 29, BARI Alu 35, BARI Alu 36, BARI Alu 37, BARI Alu 7 (Diamant), BARI Alu 8 (Cardinal) and which were characterized by lowest plant height and average values for other characters. Cluster IV contained three potato varieties BARI Alu 41, BARI Alu 46 and BARI Alu 53 which was characterized by highest plant height and second highest leaf area.

Abstract:
The study was conducted in the 20 upazila of 10 selected major potato growing districts of Bangladesh to find out the present potato cultivation status, challenges, diversity of pests and their risks management options among the farmers. The data were collected through interview of 500 potato farmers considering 25 potato farmers from each upazila. Infections of diseases were reported to be the major problems followed by lower market price of the produced potato. Other major problems faced during potato cultivation were insect pest attack and weed attack. The BARI Alu-7 (Diamant) was most susceptible potato variety to insect pests and diseases, whereas the Lal-pakhri was the least susceptible to pests. Most of the fields (98.50%) were infested by cutworm. Among these pests, cutworm and aphid were identified as major pests and caused high and moderate infestations respectively. Others were identified as minor pests of potato with low infestations. Potato tuber worm was found as the key pest in storage condition. Rat was reported as the most infesting vertebrate storage pests. Currently cutworm and aphid were more damaging insect pest of potato in field condition than previous infestation, and potato tuber worm was more damaging insect pest in storage. Insecticide spray is the most favorable pest control method among the potato growers. Only a little portion of farmers have knowledge about integrated pest management (IPM) practices. Abstract:
Random Amplified Polymorphic DNA (RAPD) markers were used to study the molecular diversity of 12 popular potato varieties in Bangladesh. DNA was extracted from tender leaf sample for PCR amplification. The PCR amplified DNA profile was visualized on 2% agarose gel, staining with ethidium bromide. Eight RAPD primers were used to evaluate the genetic diversity of potato varieties. Some total of 36 DNA fragments were amplified and out of them 24 were polymorphic. Those primers generated 61.53% of polymorphic DNA band. The primer OPX 04 produced highest (9) number of DNA band and out of 9 amplicon 6 were polymorphic. Lowest number of amplification was observed in the primer OPA-17 and it was only 3. The highest Nei’s genetic distance (0.9701) was noticed between the variety Granola and Provinto. The highest (0.8205) number of genetic identity/similarity was observed between the varieties Cardinal and Diamant. The unweighted pair group method of arithmetic mean (UPGMA) dendrogram based on Nei’s genetic distance revealed that the 12 varieties followed into two main clusters. The present finding showed that there was high level of genetic diversity among the varieties which can be used for parental selection in potato breeding program.
