Glucose oxidase production using a medicinal plant: Inula viscosa and optimization with Taguchi DOE

Abstract
The aim of the current study was to research the ability of Rhodotorula glutinis to produce glucose oxidase (GOX) enzyme and to optimize the influential environmental factors using Inula viscosa (L.) Aiton as sole carbon source. I. viscosa (L.) Aiton is an effective medicinal plant and has been widely used in folk medicine. Taguchi design of experiment (DOE) was utilized for the optimization process. The effects of I. viscosa (L.) Aiton, initial pH, incubation temperature and time were investigated at four levels using L16 orthogonal array layout of Taguchi DOE. The results showed that the most effective factor was obtained as I. viscosa (L.) Aiton and optimized conditions gained approximately 2,4 fold higher GOX activity than unoptimized conditions. Consequently, I. viscosa (L.) Aiton has a great potential for GOX production by R. glutinis and Taguchi DOE can be used as an effective optimization tool of choice for microbial enzyme production.Keywords
